Abstract

In this study, chitin and benzoic acid were conjugated by chemical methods to improve its solubility and bioactivity. The synthesis and conjugation of the chitin benzoate confirmed by FT-IR and 1 H-NMR spectra. Chitin benzoate induced cell death was further characterized by cell viability, changes in nuclear morphology and cell morphology. Significant inhibition on the cell proliferation of HeLa (human cervical adenocarcinoma) cells was observed by the effect of chitin benzoates in a dose-dependent manner. Moreover, protein and gene expression levels of the apoptosis pathway regulators such as caspase-8, Fas, FasL and Fasassociated death domain (FADD) were examined using RT-PCR and Western blot analyses. The exposure of synthesized chitin benzoate induced the apoptotic mechanism in HeLa cells via the stimulation of extrinsic pathway receptors. Therefore, the present results suggest that chitin benzoates have promising potential to use them as valuable anticancer agents.
